目的:明确鳞癌感染人类疱疹病毒8型(HV-8)ORF75基因亚型分类并初步探讨其与鳞癌的相关性。方法:对40例皮肤鳞癌、46例食管鳞癌石蜡包埋组织进行HHV-8 DNA抽提、扩增、双向测序,使用DNASTAR软件、Crustal W软件和PHYLIP软件包对测序结果进行系统发生学分析,从而确定HHV-8 ORF75基因亚型,最后运用Fisher确切概率法对结果进行统计学分析。结果:①40例皮肤鳞癌有10例HHV-8感染为阳性,阳性率为25.0%;46例食管鳞癌有12例为阳性,阳性率为26.1%.②皮肤鳞癌感染HHV-8 ORF75亚型6例为A亚型,4例为C亚型;食管鳞癌感染HHV-8 ORF75亚型8例为A亚型,4例为C亚型③鳞癌感染HHV-8 ORF75基因亚型与鳞癌患者感染部位及病理分级之间没有统计学意义(P>0.05)。结论:鳞癌患者感染HHV-8 ORF75亚型属于A亚型和C亚型;鳞癌感染HHV-8 ORF75基因亚型与鳞癌患者感染部位及病理分级无关。
OBJECTIVE: To identify the subtypes of squamous cell carcinoma of the human genital herpesvirus type 8 (HV-8) ORF75 and to explore its relationship with squamous cell carcinoma. Methods: Forty cases of squamous cell carcinoma and 46 cases of esophageal squamous cell carcinoma were subjected to HHV-8 DNA extraction, amplification and bidirectional sequencing. The phylogenetic analysis was performed using DNASTAR software, Crustal W software and PHYLIP software package Analysis, so as to determine the HHV-8 ORF75 gene subtypes, and finally use Fisher exact probability method for statistical analysis of the results. Results: ①The positive rate of HHV-8 infection in 40 cases of squamous cell carcinoma was 25.0%, 12 of 46 cases of esophageal squamous cell carcinoma were positive, the positive rate was 26.1% .②The positive rate of HHV-8 ORF75 6 cases of subtype A and 4 cases of subtype C; 8 cases of HHV-8 ORF75 subtype esophageal squamous cell carcinoma were subtypes A and 4 subtype C; There was no significant difference between the squamous cell carcinoma and the pathological grade (P> 0.05). CONCLUSIONS: The HHV-8 ORF75 subtypes in patients with squamous cell carcinoma belong to subtype A and subtype C. The ORF75 gene subtypes in squamous cell carcinoma infected HHV-8 have nothing to do with the location and pathological grade of squamous cell carcinoma.
