目的为验证碱性成纤维细胞生长因子(bFGF)对爆震性耳聋的防治作用。方法选用豚鼠35只,分为4组:bFCF肌注治疗组(10只),震后即刻给予bFGF肌注(50IU/100g)连用4周,生理盐水肌注治疗组(10只),疗程同前;bFGF肌注预防组(10只),生理盐水肌注预防组(5只),剂量同治疗组连用2周。动物爆震100发,每秒1发,脉宽0.5ms,压力峰值172.0dBSPL,爆震前后测定ABR阈值。最后处死动物做耳蜗SDH染色铺片,观察耳蜗损伤情况。结果震前肌注bFGF预防组动物阈移比生理盐水组要轻,但统计学差异不明显。耳蜗病损区域集中于一回上至二回下,细胞界线不清,SDH染色变浅。震后bFGF肌注治疗组动物震后阈移恢复较其它组快,铺片见耳蜗病变为外毛细胞节段性缺失,散在性内毛细胞缺失,病变范围从基底膜钩端至第三回下半。结论震前肌注bFGF对预防耳蜗病变从形态学上观察有一定作用;动物震后即刻bFGF肌注治疗对阈移恢复有明显作用。
Objective To verify the basic fibroblast growth factor (bFGF) on detonation induced deafness prevention and treatment. Methods 35 guinea pigs were selected and divided into 4 groups: bFCF intramuscular injection group (n = 10). Immediately after the shock, bFGF intramuscular injection (50IU / 100g) was given for 4 weeks. Normal saline intramuscular injection group Pre-bFGF group (n = 10) and saline intramuscular injection group (n = 5). The dosage was the same as that of the treatment group for 2 weeks. Animal detonation 100 rounds, 1 round per second, pulse width 0.5ms, pressure peak 172.0dBSPL, ABR threshold before and after knocking. Finally, the animals were sacrificed SDH stained cochlea to observe the damage of the cochlea. Results Before the earthquake, bFGF group was lighter than normal saline group, but the difference was not significant. Cochlear lesions focus on the back to the second back, the cell line unclear, SDH staining lighter. After shock, the bFGF intramuscular injection treatment group recovered faster than the other groups, and the cochlear lesions showed segmental loss of outer hair cells and scattered inner hair cells. The lesions ranged from the base of the basal membrane to the third Lower half. Conclusion The intramuscular injection of bFGF prior to the earthquake had a role in the prevention of cochlear lesions from morphological observation. Immediately after the bFGF injection, the intramuscular injection of bFGF significantly restored the threshold shift recovery.
