目的:了解和探讨巨噬细胞集落刺激因子(MCSF)及其受体对喉鳞状细胞癌细胞株(Hep2)细胞系的作用。方法:使用氚标记胸腺嘧啶核苷(3HTDR)掺入法和MTT法测试MCSF作用于Hep2后细胞的增殖状况,并测试加入巨噬细胞集落刺激因子受体(MC-SFR)抗体后细胞生长情况。ELISA法测试M-CSF作用Hep2细胞后上清中的MCSF的浓度变化。结果:Hep2细胞培养后较培养前的上清中的MCSF的浓度升高。加入的M-CSF浓度越大,细胞增殖越差,加入MCSFR抗体浓度越大细胞增殖越好。结论:MCSF对喉癌细胞Hep2细胞的增殖具有抑制作用。
Objective: To investigate and explore the role of macrophage colony-stimulating factor (MCSF) and its receptor on laryngeal squamous cell carcinoma cell line (Hep2). Methods: Proliferation of MCSF cells treated with MCSF was measured by 3H-TdR incorporation and MTT assay, and the cell growth was tested by adding MCSF antibody . The concentration of MCSF in the supernatant of Hep2 cells treated with M-CSF was measured by ELISA. Results: After cultured Hep2 cells, the concentration of MCSF in the supernatant before culture increased. The higher concentration of M-CSF added, the poorer the cell proliferation, the better the cell proliferation was when the concentration of MCSFR antibody was added. Conclusion: MCSF can inhibit the proliferation of laryngeal carcinoma Hep2 cells.
