目的:评价葛根芩连微丸体外干预对人轮状病毒A组G3型709株的药理作用。方法:通过细胞病变效应、噻唑蓝比色法检测细胞活性,观察葛根芩连微丸抗人轮状病毒709株的作用。结果:葛根芩连微丸的恒河猴胚胎肾细胞半数存活浓度为0.703mg/mL,对人轮状病毒709株无直接灭活杀伤作用;对人轮状病毒709株有一定预防吸附的作用,IC50=0.146mg/mL,TI=4.815068;能够抑制人轮状病毒709株细胞内生物合成的作用,IC50=0.023mg/mL,TI=30.56522。结论:葛根芩连微丸对轮状病毒吸附宿主细胞有一定干预作用,但是主要通过抑制轮状病毒在恒河猴胚胎肾细胞内的生物合成过程发挥体外抗轮状病毒作用。
Objective: To evaluate the pharmacological effects of Gegen Qinlian Pellets in vitro on human rotavirus A group G3 strain 709. Methods: The cell activity was detected by cytopathic effect (MTT) and the cell viability was assayed by thiazolyl blue colorimetry. The effect of Gegen Qinlian Pellets on the anti-human rotavirus 709 was observed. Results: Gegen Qinlian pellets of Rhesus monkey embryonic kidney cell half-live concentration of 0.703mg / mL, no direct inactivation of human rotavirus 709 killing effect; against human rotavirus 709 have some preventive effect of adsorption , IC50 = 0.146 mg / mL, TI = 4.815068; the ability to inhibit the intracellular biosynthesis of human rotavirus strain 709 was IC50 = 0.023 mg / mL and TI = 30.56522. CONCLUSION: Gegen Qinlian Pellet has some intervention effects on rotavirus host cells, but it exerts in vitro anti-rotavirus effect by inhibiting the biosynthesis of rotavirus in rhesus monkey embryonic kidney cells.
