目的:通过研究苦参素对裸鼠食管癌移植瘤组织中内质网凋亡途经相关蛋白表达的影响,探讨其诱导凋亡的相关机制。方法:通过移植瘤体积判定肿瘤生长抑制,运用TUNEL检测细胞凋亡率,采用蛋白免疫印迹技术检测内质网凋亡途径相关蛋白:结合免疫球蛋白(BIP)和C/EBP环磷酸腺苷反应元件结合转录因子同源蛋白(CHOP)的表达水平。结果:与阴性对照组比较,自给药17d起,各剂量组肿瘤体积均显著缩小(P<0.05,P<0.01);TUNEL检测中、高剂量组呈现明显细胞凋亡现象(P<0.05);中、高剂量组BIP表达显著下降(P<0.05),CHOP表达显著增加(P<0.05)。结论:苦参素注射液具有诱导裸鼠食管癌移植瘤凋亡的作用,其作用机制可能与内质网凋亡途径相关蛋白BIP的表达下调和CHOP表达上调有关。
OBJECTIVE: To investigate the effect of oxymatrine on the expression of apoptosis-related proteins in the endoplasmic reticulum of esophageal cancer xenografts in nude mice and to explore the mechanism of its apoptosis induction. Methods: Tumor growth inhibition was determined by the size of transplanted tumor. The apoptosis rate was detected by TUNEL. Western blotting was used to detect the endoplasmic reticulum apoptosis-related proteins: binding immunoglobulin (BIP) and C / EBP cyclic adenosine monophosphate Element-binding transcription factor homologous protein (CHOP) expression levels. Results: Compared with the negative control group, the tumor volume of each dose group was significantly reduced from the 17th day after administration (P <0.05, P <0.01). The apoptosis of the medium and high dose TUNEL groups showed obvious apoptosis (P <0.05). BIP expression was significantly decreased in medium and high dose groups (P <0.05), and CHOP expression was significantly increased (P <0.05). CONCLUSION: Oxymatrine injection can induce the apoptosis of transplanted esophageal cancer in nude mice. Its mechanism may be related to the down-regulation of protein BIP and the up-regulation of CHOP in endoplasmic reticulum.
