目的:从分子水平鉴别冬虫夏草与其混淆品北虫草。方法:从冬虫夏草及其常见混淆品北虫草中提取DNA,以核基因组通用引物为引物进行扩增,扩增产物经纯化后,用PCR产物直接测序法进行测序。结果:测得各样品rDNA的ITS及5.8S rDNA完全序列,18S和28S rDNA部分序列,分别为冬虫夏草459bp、北虫草548bp;二者ITS具显著差异,其中ITS1差异性达33.31%、ITS2差异性达26.67%。结论:ITS序列可有效地鉴别冬虫夏草及其混淆品北虫草。
Objective: To identify Cordyceps sinensis and its adulterants Cordyceps militaris from the molecular level. METHODS: DNA was extracted from Cordyceps militaris and its common contaminant Cordyceps militaris. The universal primers for nuclear genome were used as primers for amplification. The amplified products were purified and sequenced by direct PCR product sequencing. Results: The complete sequences of ITS and 5.8S rDNA of rDNA of each sample were determined. The partial sequences of 18S and 28S rDNA were 459bp for Cordyceps sinensis and 548bp for Cordyceps militaris, respectively. There were significant differences in ITS between the two groups, among which ITS1 difference was 33.31% and ITS2 was different. It reached 26.67%. Conclusion: ITS sequences can effectively identify Cordyceps sinensis and its adulterants, Cordyceps militaris.
