采用茎尖分生组织培养技术,获得了百合(Liliumbrownii F.E.Brown var.Viridulum Baker)的无病毒试管苗。通过基本培养基和激素配比试验,筛选出最佳的培养基组成,进行脱病毒苗的快速繁殖,并对试管苗进行生理生化指标及生物量的测试。结果表明:诱导丛生芽的最适培养基为:MS+BA(6-苄氨基嘌呤)1.0 mg/L+IAA(吲哚乙酸)0.2 mg/L+PP333(多效唑)4.0 mg/L+KT(激动素)1.0 mg/L,技术上达到了快速繁殖规模生产的要求。诱导试管苗最适生根的最适培养基为1/2 MS+IBA(鹅膏蕈氨酸)0.5 mg/L。通过电子显微镜的反复检测,筛选出10个彻底脱病毒的品种,这些品种可以为今后的无病毒优质种苗提供原种苗。发现检测的脱病毒百合试管苗的蛋白含量升高,SOD酶活升高,APX酶活升高,鲜重升高,预示脱病毒材料可能具有较好的生长势,对提高百合的产量有利。
Virus-free tube seedlings of Liliumbrownii F.E.Brown var. Viridulum Baker were obtained using shoot apical meristem culture. Through the basic culture medium and hormone ratio test, the best medium composition was screened, the rapid propagation of virus-free vaccine was carried out, and the physiological and biochemical indexes and biomass of test-tube seedlings were tested. The results showed that the optimal culture medium for inducing shoots was MS + BA (1.0 mg / L) and IAA 0.2 mg / L + PP333 4.0 mg / L + KT Kinetin) 1.0 mg / L, technically reached the requirements of rapid breeding scale production. The optimal medium for inducing the optimal rooting of test-tube plantlets was 1/2 MS + IBA (ibotenic acid) 0.5 mg / L. Repeated detection by electron microscopy, screening out 10 completely virus-free varieties, these varieties can provide the original seedless vaccine-free high-quality seedlings. It was found that the content of the tested virus-free lily in vitro increased, the activity of SOD increased, the activity of APX increased, the fresh weight increased, indicating that the virus-free material may have good growth potential, which is beneficial to increase the yield of lily.
