【目的】揭示广东汉族人群Y染色体小卫星DYF155S1基因座3’端多态性。【方法】采用Amp-FLP和荧光MVR-PCR方法扩增,PCR产物用ABI377电泳或中性聚丙烯酰胺凝胶电泳-银染方法分析。【结果】155例无关男性个体的4型核心序列MVR图谱第1条DNA片段大小均为133bp。155例共检出有24种等位基因,有7个等位基因仅出现1次,频率最高的为9号等位基因,DNA条带数为连续的13个,频率为0.1355;基因多样性(h)为0.9226。155人中有3人表现为连续DNA谱带中有1个DNA条带的缺失(nullrepeat),序列分析证明缺失的DNA条带位置对应于3型核心序列。【结论】揭示了DYF155S1位点3’端多态性及其等位基因频率,为群体遗传学研究及法医学应用提供了基础资料。
【Objective】 To reveal the polymorphism of DYF155S1 locus 3 ’end of Y chromosome small satellite in Guangdong Han population. 【Method】 The PCR products were amplified by Amp-FLP and fluorescence MVR-PCR. The PCR products were analyzed by ABI377 or neutral polyacrylamide gel electrophoresis-silver staining. 【Result】 The results showed that the size of the first DNA fragment in MVR map of type 4 core sequence in all 155 unrelated males was 133bp. There were 24 kinds of alleles detected in 155 cases, with 7 alleles occurring only once and the highest frequency being 9 alleles. The number of DNA bands was 13 in a row with a frequency of 0.1355. The genetic diversity (h) was 0.9226.155. Three of them showed null DNA repertoire in consecutive DNA bands. Sequence analysis showed that the deleted DNA bands corresponded to type 3 core sequences. 【Conclusion】 The 3 ’polymorphism of DYF155S1 locus and its allele frequency were revealed, which provided basic information for population genetics and forensic applications.
