目的 :为了研究川芎嗪、丹参两味中药对体外培养心肌成纤维细胞 (fibroblasts,Fbs)的胶原合成与细胞增殖的影响。方法 :采用胶原酶和胰蛋白酶消化SD大鼠的心肌组织块 ,制备成纤维细胞的单细胞悬液 ,按不同药物及药物的不同浓度分组研究它们对成纤维细胞的胶原合成和增殖影响。结果 :中、高浓度的川芎嗪、丹参能有效地抑制成纤维细胞的胶原合成和增殖。低浓度的川芎嗪、丹参能对抗去甲肾上腺素对成纤维细胞的胶原合成与细胞增殖。结论 :川芎嗪、丹参能抑制成纤维细胞的胶原合成与细胞增殖。这些效应的机制可能与它们的钙离子拮抗有关。
Objective: To study the effects of the traditional Chinese medicines Ligustrazine and Salviae miltiorrhizae on collagen synthesis and cell proliferation of cultured fibroblasts (Fbs) in vitro. METHODS: Cardiac tissue blocks from SD rats were digested with collagenase and trypsin to prepare single cell suspensions of fibroblasts. The effect of different concentrations of drugs and drugs on the collagen synthesis and proliferation of fibroblasts was studied. RESULTS: Medium and high concentrations of tetramethylpyrazine and salvia miltiorrhiza can effectively inhibit collagen synthesis and proliferation of fibroblasts. Low concentrations of tetramethylpyrazine and salvia miltiorrhiza can counteract the effects of noradrenaline on collagen synthesis and cell proliferation of fibroblasts. Conclusion : Ligustrazine and Salvia miltiorrhiza can inhibit collagen synthesis and cell proliferation in fibroblasts. The mechanism of these effects may be related to their calcium antagonism.
