目的检测人慢性粒细胞白血病K562细胞来源外泌体基质金属蛋白酶-9(matrix metalloproteinase-9,MMP9)和趋化因子受体-4(chemokine receptor 4,CXCR4)基因的表达,探讨K562细胞外泌体可能的功能。方法采用RT-PCR方法分别检测K562细胞和K562/Adr细胞(阿霉素耐药株)及两者分泌的外泌体MMP9、CXCR4基因的表达。结果在K562细胞和K562/Adr细胞中,均检测到MMP9和CXCR4基因的表达。而在K562细胞和K562/Adr细胞分泌的外泌体中,只检测到MMP9基因的表达。结论两种K562细胞都可以分泌一定量的外泌体;虽然两种K562细胞都表达MMP9、CXCR4基因,但外泌体只选择性携带和转运MMP9基因,推测其可能具有促血管新生功能。
Objective To detect the expression of matrix metalloproteinase-9 (MMP9) and chemokine receptor 4 (CXCR4) gene exosomes in human chronic myelogenous leukemia K562 cells, Body possible function. Methods The expressions of MMP9 and CXCR4 genes in K562 cells, K562 / Adr cells (doxorubicin-resistant) and exosome secreted by these two methods were detected by RT-PCR. Results The expression of MMP9 and CXCR4 genes was detected in K562 cells and K562 / Adr cells. In exosomes secreted by K562 cells and K562 / Adr cells, only the expression of the MMP9 gene was detected. Conclusions Both K562 cells can excrete a certain amount of exosomes. Although both K562 cells express MMP9 and CXCR4 genes, exosomes only selectively carry and transport MMP9 gene, suggesting that they may have pro-angiogenic functions.
