目的 :观察新霉素、苯丁酸钠及两者联合应用对胶质瘤的抑制作用。方法 :将F98胶质瘤细胞注入Fischer3 44大鼠尾状核头处制成鼠脑胶质瘤模型 (治疗组各 2 0只 ,对照组 10只 )。于种植后第 10天开始将新霉素 10mmol·L-1局部注入到肿瘤中心 ;苯丁酸钠 60mgbid ,腹腔注射。分别与治疗后第 6,12 ,18天行MRI增强扫描检查并计算肿瘤体积。结果 :治疗组 60只全部存活。前后体积对比显示治疗组肿瘤缩小显著 :新霉素组 ( 4 4 5± 1 1)mm3 相对于 ( 3 2 2± 0 9)mm3 ,P <0 0 5 ;苯丁酸钠组 ( 4 6 2± 0 9)mm3 相对于 ( 15 4± 1 1)mm3 ,P <0 0 1;联合组 ( 4 5 0± 1 1)mm3 ,相对于 ( 8 2± 2 1)mm3 ,P <0 0 1;对照组有 8只死亡 ,体积对比显示肿瘤明显增大 [( 4 5 3± 1 2 )mm3 相对于 ( 68 1± 1 2 )mm3 ,P <0 0 5 ]。结论 :新霉素和苯丁酸钠对胶质瘤细胞有抑制作用 ,联合用药抑制胶质瘤效果显著。
Objective: To observe the inhibitory effect of neomycin, sodium phenylbutyrate and their combination on glioma. METHODS: F98 glioma cells were injected into the caudate nucleus of Fischer3 44 rats to establish a rat glioma model (20 in the treatment group and 10 in the control group). On the 10th day after planting, neomycin 10 mmol·L-1 was locally injected into the tumor center; sodium phenylbutyrate 60 mg bid was injected intraperitoneally. The MRI scan and the tumor volume were performed respectively on the 6th, 12th and 18th days after treatment. Results: All 60 patients in the treatment group survived. The comparison of volume before and after treatment showed that the tumors in treatment group shrank conspicuously: neomycin group (445 ± 1 1) mm3 relative to (32 2 ± 0 9) mm3, P <0 05; sodium phenylbutyrate group ± 0 9) mm 3 relative to (15 4 ± 1 1) mm 3, P 0 01; combined group 4 54 0 ± 1 1 mm 3 vs 8 2 ± 2 1 mm 3 P 0 01 There were 8 deaths in the control group. The volume contrast showed a significant increase in the tumor size (453 ± 1 2) mm3 vs (68 1 ± 1 2) mm3, P <0 05). Conclusion: Neomycin and sodium phenylbutyrate have inhibitory effects on glioma cells, and combined therapy can inhibit the glioma effectively.
