利用~1H NMR谱研究枯草杆菌中性蛋白酶与无机金属化合物(CoCl_2, NiCl_2)间的相互作用.结果发现,原酶活性中心金属离子Zn(Ⅱ)可以与外加CoCl_2,NiCl_2发生直接相互作用,被Co(Ⅱ),Ni(Ⅱ)部分置换而生成了酶的Co(Ⅱ),Ni(Ⅱ)取代衍生物;获得了该酶的两种金属取代衍生物的~1HNMR谱图,同时论证了该酶活性中心的配位结构.
Study on the Interaction between Bacillus subtilis Neutral Protease and Inorganic Metal Compounds (CoCl_2, NiCl_2) by ~ 1H NMR Spectroscopy. The results showed that Zn (Ⅱ) could directly interact with CoCl_2 and NiCl_2, and Co (Ⅱ) and Ni (Ⅱ) replaced Co (Ⅱ) and Ni Substituted derivatives; ~ 1HNMR spectra of two metal-substituted derivatives of the enzyme were obtained, and the coordination structure of the active center of the enzyme was demonstrated.
