纳米二氧化钛对金鱼脑组织的氧化应激损伤

为探讨光催化型nano-Ti O2(纳米二氧化钛)对金鱼(Carassius auratus L.)脑组织的氧化应激损伤效应,以5 nm的锐钛型钛白粉为试验材料,分别在ρ(nano-Ti O2)为0、16、32、64、128 mgL下对金鱼进行处理,测定金鱼脑组织MDA(丙二醛)、PC(羰基化蛋白)和H2O2含量,SOD(超氧化物歧化酶)和GPx(谷胱甘肽过氧化物酶)活性以及CuZn-SOD蛋白表达量.结果表明:ρ(nano-Ti O2)从0 mgL升至128 mgL,金鱼脑组织CuZn-SOD活性从1.70×10-3katkg(以鲜质量计,下同)降至0.35×10-3katkg,再回升至0.46×10-3katkg;Mn-SOD活性呈降—升—降趋势,MDA含量从0.85 nmolg升至2.83 nmolg,之后降至2.54nmolg;PC含量一直上升;H2O2含量从4.32μmolg升至9.28μmolg,之后降至8.38μmolg.随着ρ(nano-Ti O2)的加大,CuZnSOD蛋白表达量逐渐减少.研究表明,nano-Ti O2对金鱼脑组织具有很强的氧化损伤作用,MDA与PC含量升高与CuZn-SOD和GPx活性下降相关,H2O2含量升高与GPx活性下降等因素有关. In order to investigate the effect of photocatalytic nano-Ti O2 on the oxidative stress in the brain of goldfish (Carassius auratus L.), anatase titanium dioxide (5 nm) O2) at 0, 16, 32, 64 and 128 mg  L, respectively. The contents of malondialdehyde (MDA), PC (carbonylation protein) and H2O2 in the brain of goldfish were determined, and the activities of SOD (superoxide dismutase) And GPx (glutathione peroxidase) activity and the expression of CuZn-SOD protein were measured.The results showed that the content of ρ (nano-Ti O2) increased from 0 mgL to 128 mgL, Zn-SOD activity decreased from 1.70 × 10-3katkg (fresh weight, the same below) to 0.35 × 10-3katkg, and then rose to 0.46 × 10-3katkg; Mn-SOD activity decreased from - decreased, the content of MDA increased from 0.85 nmolg to 2.83 nmolg, then decreased to 2.54nmolg; the content of PC increased all the time; the content of H2O2 increased from 4.32μmolg to 9.28μmolg, then decreased to 8.38μmol g.With the increase of ρ (nano-Ti O2), the expression of CuZnSOD protein gradually decreased.The results showed that nano-Ti O2 had a strong oxidative damage on the brain of goldfish, and the content of MDA and PC were increased Decreasing phase with CuZn-SOD and GPx activity Guan, H2O2 content and GPx activity decreased and other factors.
