目的探讨膜-细胞骨架结合磷酸化蛋白50基因(ezrin-radixin-moesin-binding phosphoprotein 50,EBP50)与乳腺癌特异性基因1(breat cancer-specific gene 1,BCSG1)在子宫内膜癌中的表达及相互关系。方法子宫内膜癌患者67例为子宫内膜癌组,子宫内膜非典型增生患者68例为增生组,子宫内膜正常患者70例为对照组,分别取3组患者的子宫内膜癌组织、非典型增生组织和子宫内膜正常组织,采用免疫组织化学法检测3组EBP50及BCSG1蛋白表达水平,采用荧光定量PCR检测EBP50及BCSG1mRAN表达水平。结果子宫内膜癌组EBP50和BCSG1mRNA表达水平(2.58±1.20、0.84±0.10)以及其蛋白阳性率(88.05%、79.10%)均高于增生组(1.76±0.53、0.58±0.14和35.29%、27.94%)及对照组(1.06±0.65、0.21±0.20和12.85%、5.71%)(P<0.05),增生组高于对照组(P<0.05);子宫内膜癌组EBP50mRAN表达与BCSG1mRAN表达呈正相关(r=0.725,P=0.001)。结论 EBP50和BCSG1基因均与子宫内膜癌的发生、发展有关,EBP50和BCSG1对子宫内膜癌的发生和调控有协同作用。
Objective To investigate the expression of membrane-cytoskeleton-associated phosphoprotein 50 (EBP50) and the expression of the bladder cancer-specific gene 1 (BCSG1) in endometrial carcinoma And interrelationships. Methods 67 patients with endometrial cancer were endometrial cancer group, 68 patients with endometrial dysplasia were hyperplastic group, 70 patients with normal endometrium were control group, three groups of patients with endometrial cancer tissue , Atypical hyperplasia tissues and endometrial normal tissues. The expressions of EBP50 and BCSG1 protein were detected by immunohistochemical method. The expression levels of EBP50 and BCSG1mRAN were detected by real-time PCR. Results The expression levels of EBP50 and BCSG1 mRNA in endometrial carcinoma group (2.58 ± 1.20,0.84 ± 0.10) and their protein positive rate (88.05%, 79.10%) were significantly higher than those in proliferative group (1.76 ± 0.53,0.58 ± 0.14 and 35.29%, 27.94 %) And the control group (1.06 ± 0.65,0.21 ± 0.20 and 12.85%, 5.71%, respectively) (P <0.05). The expression of EBP50mRAN in endometrial carcinoma was positively correlated with the expression of BCSG1mRAN (r = 0.725, P = 0.001). Conclusion Both EBP50 and BCSG1 genes are associated with the occurrence and development of endometrial carcinoma. EBP50 and BCSG1 have synergistic effects on the occurrence and regulation of endometrial carcinoma.
