目的 比较不同培养条件下原代神经球细胞中三种多巴胺受体亚型表达的变化 ,探讨发育过程中不同亚型多巴胺受体表达调控的机制。方法 流产胎儿脑组织经体外培养获得神经干细胞球 ,再用各种因子进行诱导分化 ,提取细胞总RNA并用RT -PCR方法检测mRNA转录。结果 在神经球细胞中三种多巴胺受体均有表达 ,中脑与海马来源的神经球细胞无显著差异。在无血清培养的条件下 ,表皮生长因子 (EGF) ,碱性成纤维细胞生长因子 (bFGF)及白介素 (Ils)诱导均使得D3 受体表达下调 ,而有血清培养时 ,白介素 (Ils)诱导则不能出现下调或关闭的结果。结论 因子诱导产生的分化对于D3 受体表达起下调或关闭作用。
Objective To compare the expression changes of three dopamine receptor subtypes in primary neurospheres under different culture conditions and to explore the mechanism of dopamine receptor expression in different subtypes during the development. Methods Aborted fetal brain tissue was harvested from neural stem cells in vitro and induced by various factors. Total RNA was extracted and mRNA transcription was detected by RT-PCR. Results The expression of all three dopamine receptors in neurosphere cells was not significantly different between neuroblasts derived from midbrain and hippocampus. Under the condition of serum-free culture, the expression of D3 receptor was down-regulated by the induction of epidermal growth factor (EGF), basic fibroblast growth factor (bFGF) and interleukin (Ils), while IL- You can not appear the result of the down or closed. Conclusion Differentiation induced by factor-induced down-regulation or down-regulation of D3 receptor expression.
