目的检测缺氧诱导因子-1α(HIF-1α)在酒精性急性胃黏膜损伤中的表达,探讨其在酒精性急性胃黏膜损伤中的作用。方法 60只SD大鼠随机分为4组(每组15只):对照组和Ⅰ、Ⅱ、Ⅲ实验组。对照组以2ml生理盐水灌胃,Ⅰ、Ⅱ、Ⅲ实验组分别予30%、65%、100%浓度酒精2ml灌胃。2h后处死大鼠取全胃剖开,肉眼观察形态学变化计算损伤指数;HE染色后光镜观察黏膜炎症程度;RT-PCR检测胃黏膜HIF-1αmRNA表达,蛋白免疫切迹法检测HIF-1α蛋白表达。结果肉眼观察实验组黏膜有充血、水肿及糜烂、溃疡形成,光镜下观察实验组均有炎症反应;对照组与实验Ⅰ、Ⅱ、Ⅲ组损伤指数分别为0.47±0.52、9.27±2.49、11.07±2.71、28.60±3.11,实验各组损伤指数与对照组相比差异有统计学意义(P均<0.05),且随着酒精浓度增加损伤更为严重;实验组HIF-1αmRNA及蛋白表达随酒精浓度升高而递增(P<0.01或P<0.05)。结论随着酒精浓度的增高,大鼠急性胃黏膜损伤的程度加重,且胃黏膜HIF-1α的表达水平增加,推测HIF-1α在酒精性胃黏膜损伤过程中起重要作用。
Objective To investigate the expression of hypoxia inducible factor-1α (HIF-1α) in alcoholic acute gastric mucosal injury and its role in alcoholic acute gastric mucosal injury. Methods Sixty SD rats were randomly divided into 4 groups (n = 15): control group and experimental group Ⅰ, Ⅱ, Ⅲ. The control group was given gavage with 2ml normal saline, and the experimental group with Ⅰ, Ⅱ and Ⅲ was treated with 2ml alcohol by 30%, 65% and 100% respectively. After 2h, the rats were sacrificed and the whole stomach was cut open. The damage index was calculated by macroscopic observation. The degree of mucosal inflammation was observed by light microscopy after HE staining. The expression of HIF-1α mRNA in gastric mucosa was detected by RT-PCR and HIF-1α Protein. Results The mucosa of the experimental group were hyperemic, edematous, erosive and ulcerated. The inflammatory reaction was observed in the experimental group under the light microscope. The damage indices of the control group and the experimental groups Ⅰ, Ⅱ and Ⅲ were 0.47 ± 0.52, 9.27 ± 2.49 and 11.07 ± 2.71,28.60 ± 3.11. There was significant difference in injury index between experimental group and control group (P <0.05), and more serious with the increase of alcohol concentration. The expression of HIF-1αmRNA and protein in experimental group were significantly decreased with alcohol Concentration increased with increasing (P <0.01 or P <0.05). Conclusions With the increase of alcohol concentration, the degree of acute gastric mucosal lesion in rats increases and the expression of HIF-1α in gastric mucosa increases. It is speculated that HIF-1α plays an important role in alcoholic gastric mucosal injury.
