目的探讨DNA甲基化转移酶(DNMTs)与急性A型主动脉夹层(Acute aortic dissection,AAD)发生发展之间的关系。方法收集5例急性A型AAD患者病变血管组织和4例无主动脉疾病的器官捐献者的升主动脉血管组织,采用Western Blot方法检测DNMTs(DNMT1、DNMT3A、DNMT3B、DNMT3L)的蛋白在患者和正常对照血管组织中的表达情况。结果与健康对照组相比,整体上DNA甲基化转移酶蛋白在A型AAD患者病变血管组织中表达下降,DNMT1蛋白在A型AAD患者病变血管组织中表达较正常对照下降约50%(P=0.023),DNMT3B蛋白在A型AAD患者病变血管组织中表达下降约50%(P=0.009);DNMT3A和DNMT3L蛋白在A型AAD患者病变血管组织中表达下降趋势,尽管无统计学意义(P=0.414,P=0.16)。结论 DNA甲基化转移酶在急性A型AAD患者病变血管组织中表达水平较正常对照呈下降趋势,DNA甲基化是否参与A型AAD病变的发生发展尚需进一步研究。
Objective To investigate the relationship between DNA methylation transferase (DNMTs) and the development of Acute aortic dissection (AAD). Methods Aortic vascular tissue was collected from 5 patients with acute type A AAD and 4 donor organs without aortic disease. The protein levels of DNMTs (DNMT1, DNMT3A, DNMT3B and DNMT3L) were detected by Western Blot in patients and Normal control vascular tissue expression. Results Compared with the healthy control group, the expression of DNA methylation transferase decreased in the diseased vascular tissue of type A AAD patients and DNMT1 protein decreased in the diseased vascular tissue of type A AAD patients by about 50% (P = 0.023). The DNMT3B protein expression was decreased by about 50% (P = 0.009) in the diseased vascular tissue of type A AAD patients. The expression of DNMT3A and DNMT3L protein in the diseased vascular tissue of type A AAD patients was decreased, although not statistically significant = 0.414, P = 0.16). Conclusions The expression of DNA methylase in patients with acute A-type AAD is lower than that in normal controls. Whether DNA methylation is involved in the development of A-type AAD remains to be further studied.
