目的:观察益气活血复方对mt TFA在心力衰竭大鼠中表达的影响。方法:大鼠80只,随机选取20只为正常组,余大鼠经左冠状动脉前降支结扎法配合力竭式游泳、节食等方法造成慢性心衰模型,将造模成功大鼠分为模型组、中药组、西药组,共4组,均以灌胃给药或蒸馏水28 d。利用蛋白免疫印迹法和RT-PCR法分别检测线粒体转录因子A(mitochondrial transcription factor A,mt TFA)的蛋白和基因表达的变化情况,比色法测定高能磷酸盐(adenosine triphosphate,ATP)浓度,ELISA法测定血清脑钠素(brain natriuretic peptide,BNP)的浓度。结果:与正常组相比,模型组及余各组大鼠ATP及mt TFA水平显著降低(P<0.01),BNP含量显著升高(P<0.01);与模型组相比,中药组和西药组ATP及mt TFA明显升高(P<0.01),BNP含量明显降低(P<0.01);西药组与中药组血清中ATP,mt TFA及BNP含量无明显差异(P>0.05)。结论:益气活血中药可显著降低心衰大鼠BNP的含量水平,改善线粒体能量代谢,改善心功能,促进mt TFA生成,可抑制或逆转心室重构的过程,从而有抗心力衰竭的作用。
Objective: To observe the effect of Yiqi Huoxue Compound on the expression of mt TFA in heart failure rats. Methods: Totally 80 rats were randomly selected and 20 rats were selected as the normal group. Chronic heart failure model was induced by left anterior descending coronary artery ligation combined with exhaustive swimming and diet. The rats were divided into Model group, Chinese medicine group, Western medicine group, a total of 4 groups, were intragastrically administered or distilled water 28 d. The changes of protein and gene expression of mitochondrial transcription factor A (mt TFA) were detected by Western blotting and RT-PCR respectively. The concentrations of adenosine triphosphate (ATP), ELISA Method to determine the concentration of brain natriuretic peptide (BNP). Results: Compared with the normal group, the levels of ATP and mt TFA in the model group and the rest groups were significantly decreased (P <0.01) and BNP content was significantly increased (P <0.01). Compared with the model group, The contents of ATP, mt TFA and BNP were significantly increased (P <0.01) and BNP content was significantly lower in the two groups than those in the control group (P <0.01). Conclusion: Yiqi Huoxue Chinese medicine can significantly reduce BNP levels in heart failure rats, improve mitochondrial energy metabolism, improve cardiac function, promote the production of mt TFA, inhibit or reverse the process of ventricular remodeling, and thus have anti-heart failure effect.
