目的探讨小蘖碱对体外培养人卵巢癌SKOV-3细胞增殖和凋亡的影响。方法选用人卵巢癌细胞株SKOV-3进行体外培养,以5-100μmol/L小蘖碱分别处理SKOV-3细胞24-72h,MTT法检测细胞的生长抑制率;流式细胞仪检测细胞周期;SABC法检测survivin、caspase-3在细胞中的表达。结果小蘖碱能显著抑制SKOV-3细胞体外生长,呈时间剂量依赖性。小蘖碱作用24h随浓度的升高survivin阳性表达逐渐降低,caspase-3阳性表达逐渐升高。流式细胞仪分析证实小蘖碱能使SKOV-3细胞积聚在S期和G2/M期。结论小蘖碱对人卵巢癌细胞株SKOV-3的生长有显著的抑制作用,survivin的表达下降、caspase-3的表达上调可能参与了诱导细胞凋亡。
Objective To investigate the effects of berberine on the proliferation and apoptosis of human ovarian cancer cell line SKOV-3 in vitro. Methods Human ovarian cancer cell line SKOV-3 was cultured in vitro. SKOV-3 cells were treated with 5-100μmol / L berberine for 24-72h. The cell growth inhibition rate was determined by MTT assay. Cell cycle was detected by flow cytometry. SABC method was used to detect the expression of survivin and caspase-3 in cells. Results Berberine could significantly inhibit the growth of SKOV-3 cells in vitro in a time-and dose-dependent manner. With the concentration of berberine 24h, the expression of survivin gradually decreased and the expression of caspase-3 gradually increased. Flow cytometry analysis confirmed that berberine could accumulate SKOV-3 cells in S phase and G2 / M phase. Conclusion Berberine can significantly inhibit the growth of human ovarian cancer cell line SKOV-3, and the expression of survivin is down-regulated. The up-regulation of caspase-3 may be involved in the induction of apoptosis.
