目的:研究培养雪莲总黄酮对炎性介质一氧化氮(NO),前列腺素E2(PGE2)分泌以及一氧化氮合酶(iNOS),环氧酶-2(COX-2)mRNA表达的影响,探讨其抗炎机制。方法:用Griess法测定培养雪莲总黄酮对Raw264.7细胞NO的影响,放射免疫法测定培养雪莲总黄酮对Raw264.7细胞PGE2含量;逆转录聚合酶链式反应方法检测Raw264.7细胞iNOS,COX-2mRNA表达情况。结果:培养雪莲总黄酮可明显降低Raw264.7细胞上清中NO,PGE2含量,同时,降低iNOS,COX-2 mRNA表达。结论:培养雪莲总黄酮的抗炎作用与抑制炎性介质NO,PGE2分泌有关,抑制iNOS,COX-2 mRNA表达从而降低炎性介质NO,PGE2分泌可能是培养雪莲总黄酮的抗炎作用的机制之一。
Objective: To study the effects of total flavonoids of Radix Eua Xu on expression of nitric oxide (NO), prostaglandin E2 (PGE2), and expression of iNOS and COX-2 mRNA in inflammatory mediators, Explore its anti-inflammatory mechanism. Methods: The content of NO in raw264.7 cells was determined by Griess method. The content of PGE2 in raw Rawl64.7 cells was determined by radioimmunoassay. The iNOS expression in Raw264.7 cells was detected by reverse transcription-polymerase chain reaction (RT-PCR) COX-2 mRNA expression. Results: Cultivation of total flavonoids in Elaeocarpus sylvestris L. could significantly reduce the content of NO and PGE2 in the supernatant of Raw264.7 cells and decrease the expressions of iNOS and COX-2 mRNA. CONCLUSION: The anti-inflammatory effect of total flavonoids from Saxifolium japonicus is related to inhibiting the secretion of NO and PGE2, inhibiting the expression of iNOS and COX-2 mRNA and decreasing the secretion of NO and PGE2 in inflammatory mediators may be the mechanism of anti-inflammatory effects of total flavones one.
