本文报道了一种同时适用于水稻种子和叶片的简单快速DNA提取方法。该方法通过简单的碾磨、加热、离心等步骤便能制备适合PCR的DNA模板。用该方法单人每小时可以提取48个样品的DNA模板。本研究用该方法提取了132个水稻种子DNA样品及1096个不同生长阶段的水稻叶片DNA样品,用13对PCR引物进行扩增,种子DNA样品PCR扩增成功率为91.0%,3个不同时期取材的叶片DNA样品PCR扩增成功率均在97.0%以上。该方法提取的DNA也可以进行高分辨率熔解曲线分析。
This paper reports a simple and rapid DNA extraction method that is suitable for both rice seeds and leaves. This method can be used to prepare DNA template suitable for PCR by simple milling, heating, centrifugation and other steps. With this method, 48 DNA samples per hour can be extracted from the sample. In this study, DNA samples from 132 rice seeds and 1096 rice leaf samples from different growth stages were extracted by this method. 13 pairs of PCR primers were used to amplify DNA samples. The success rate of DNA amplification was 91.0% The successful rate of PCR amplification of the sampled DNA samples was more than 97.0%. The DNA extracted by this method can also be used for high-resolution melting curve analysis.
