目的探讨雪胆甲素(CucurbitacinⅡa)对人非小细胞肺癌(Non-small cell lung cancer,NSCLC)A549细胞增殖的影响。方法以不同浓度的雪胆甲素作用于A549细胞和小鼠原代培养的脾细胞,采用MTT法检测雪胆甲素对两种细胞增殖的影响;流式细胞术及Annexin V-PE细胞凋亡试剂盒检测雪胆甲素对A549细胞细胞周期及凋亡的影响。结果雪胆甲素对A549细胞和小鼠原代脾细胞的增殖均有抑制作用,且呈浓度依赖性,25~100μmol/L的雪胆甲素对A549细胞的增殖抑制率明显高于脾细胞;雪胆甲素可将大部分A549细胞阻滞于G0/G1和S期,少部分细胞阻于G2和M期,并可促进细胞凋亡。结论雪胆甲素可明显抑制A549细胞增殖,在NSCLC高效低毒的治疗药物的开发中具有重要价值。
Objective To investigate the effects of Cucurbitacin Ⅱa on the proliferation of human non-small cell lung cancer (NSCLC) A549 cells. Methods Different concentrations of snow hyacinth A were applied to A549 cells and mouse primary cultured splenic cells. MTT assay was used to detect the effects of chloradyalcin on the proliferation of the two cell lines. Flow cytometry and Annexin V-PE cell apoptosis Effect of Serum Actinin on Cell Cycle and Apoptosis in A549 Cells. Results Serum CL had inhibitory effects on the proliferation of A549 cells and primary mouse spleen cells in a concentration-dependent manner. The inhibitory rate of CL on A549 cells was significantly higher than that of splenocytes at 25-100 μmol / L ; Acupoint A could block most of A549 cells in G0 / G1 and S phase, a small number of cells in G2 and M phase, and can promote apoptosis. Conclusion It can significantly inhibit the proliferation of A549 cells and play an important role in the development of NSCLC with high efficacy and low toxicity.
