目的 探讨胰岛素样生长因子 1(IGF I)对脐血免疫活性细胞功能的影响。方法 采用3 H TdR掺入法和3 H TdR释放法从T细胞增殖 ,NK细胞杀伤活性 ,CTL杀伤活性三个方面来探讨IGF I对脐血免疫活性细胞功能的影响。结果 (1)IGF I能明显增强PHA刺激的脐血T细胞DNA合成 ,促进其增殖 (P <0 0 0 1)。 (2 )IGF I能明显增强脐血NK细胞的基础杀伤活性和IL 2激活的NK细胞杀伤活性 (P <0 0 0 5 ) ,经IL 2和IGF I联合处理后 ,脐血NK细胞杀伤活性接近IGF I处理后的年长儿NK细胞杀伤活性 (P >0 0 5 )。 (3)IGF I能明显增强脐血CTL的杀伤活性 (P <0 0 5 )。结论 IGF I对多种脐血免疫活性细胞均有正性调节作用 ,可望将其作为一种免疫刺激剂应用于临床 ,但因人体神经内分泌免疫系统网络的复杂性 ,需作进一步研究以阐明其具体机制 ,为广泛应用于临床打下基础
Objective To investigate the effect of insulin-like growth factor 1 (IGF-I) on the function of cord blood immunocompetent cells. Methods The effects of IGF I on the function of cord blood immunocompetent cells were investigated by 3 H TdR incorporation and 3 H TdR release from three aspects: T cell proliferation, NK cell killing activity and CTL killing activity. Results (1) IGF I could significantly enhance the DNA synthesis of cord blood T cells stimulated by PHA and promote its proliferation (P <0.01). (2) IGF I could significantly enhance the basic cytotoxic activity of NK cells in cord blood and the cytotoxicity of NK cells activated by IL 2 (P <0 05). After combined treatment with IL 2 and IGF I, the cytotoxicity of NK cells in umbilical cord blood Close to the IGF I-treated NK cell cytotoxic activity (P> 0 05). (3) IGF I could significantly enhance CTL cytotoxic activity in umbilical cord blood (P <0.05). CONCLUSION IGF I has a positive regulatory effect on a variety of cord blood immunocompetent cells and is expected to be used clinically as an immunostimulant.
