万古霉素系糖肽类抗生素,临床上用于耐青霉素金葡菌所致感染。本品在肾脏功能损伤病人中血清半衰期延长,由于受体温影响,导致晶状降解产物CDP-1蓄积。在此介绍一种同时测定万古霉素和CDP-1含量的HPLC法。试剂将盐酸万古霉素贮备液加到冷藏血清中,制备5、10、20和50μg/ml万古霉素标准液。将新鲜配制盐酸CDP-1碳酸氢钠缓冲液(0.05mol/L,pHg.0)的贮备液加到冷藏血清中,制备1、2、5和10μg/mlCDP-1标准液。用水配制0.4mg/ml维生素B_(12)内标液。各标准液于-20℃贮存。仪器采用Beckman Gold HPLC系统。色谱分析采用十八硅烷柱,粒度5μm。也可使用ODS反相C_(18)柱(4.6mm×22cm)。色谱条件万古霉素和内标B_(12)的检测波长分别为235nm和360nm。采用梯度洗脱的
