:目的 :观察富硒珍珠茶对人肝癌细胞株Bel- 740 2诱导分化的影响。方法 :方法Ⅰ采用血清药理学方法 ,给家兔灌服富硒珍珠茶 7天后 ,取其血清加至细胞培养基中进行实验 ;方法Ⅱ采用直接加富硒珍珠茶提取液至细胞培养基中进行实验。应用细胞生物学方法对处理后的Bel- 740 2细胞进行生长曲线、克隆形成、AFP和ALB的合成和分泌 ,γ -GT活力的测定及细胞形态的观察。结果 :富硒珍珠茶及血清能够抑制细胞生长及克隆形成 ,减少AFP的合成和分泌 ,抑制γ-GT活性 ,促进ALB的合成和分泌 ,并呈现一定的浓度和时间依赖关系 ,促使细胞形态向正常方向分化。结论 :富硒珍珠茶能够诱导人肝癌细胞株向正常方向分化 ,具有预防原发性肝癌发生的作用
Objective : To observe the effect of selenium-enriched pearl tea on the differentiation of human hepatoma cell line Bel-740 2 . METHODS: Method I Serum pharmacological methods were used to administer selenium-enriched pearl tea to rabbits for 7 days. Then, the serum was added to the cell culture medium for experimentation. Method II was to directly add selenium-enriched pearl tea extract to the cell culture medium. conduct experiment. The growth curve, colony formation, synthesis and secretion of AFP and ALB, determination of γ-GT activity and observation of cell morphology were performed on treated Bel-740 2 cells using cell biology methods. RESULTS: Selenium-enriched pearl tea and serum could inhibit cell growth and colony formation, reduce the synthesis and secretion of AFP, inhibit γ-GT activity, promote the synthesis and secretion of ALB, and exhibit a certain concentration and time dependence, promoting cell morphology. Normal differentiation. Conclusion :Selenium-enriched pearl tea can induce differentiation of human hepatocellular carcinoma cell lines in normal direction, and it can prevent the occurrence of primary liver cancer.
