目的探讨下丘脑-脑垂体胃动素(MTL)对消化间期胃运动的调控作用。方法 32只大鼠按数字表法随机分为4组(每组8只):下丘脑室旁核(PVN)+MTL组,PVN+生理盐水(NS)组,垂体摘除+PVN+MTL组,垂体摘除+PVN+NS组。采用荧光免疫组织化学染色方法,观察大鼠脑垂体MTL免疫物质的表达;采用在体胃运动记录的方法,观察大鼠垂体摘除后血浆MTL和消化间期移行性复合运动(MMC)的改变。结果 (1)PVN+MTL组注射MTL后,MMC收缩频率显著加快,为[(11.2±1.3)次/h,PVN+NS组为[(4.9±0.6)次/h],与对照值[(5.3±0.5)次/h]比较差异无统计学意义(P>0.05);(2)垂体摘除后2 d,MMC收缩频率显著减慢[(4.5±0.8)次/h](对照值),垂体摘除+PVN+MTL组MMC收缩频率增快[(8.2±0.7)次/h],与对照值比较差异有统计学意义(P<0.05);垂体摘除+PVN+NS组[(4.3±0.9)次/h]与对照值比较差异无统计学意义(P>0.05);(3)免疫组织化学荧光研究结果显示,脑垂体组织有MTL免疫阳性反应物表达。结论下丘脑-脑垂体对消化间期胃运动有调控作用。
Objective To investigate the regulation of hypothalamic-pituitary motilin (MTL) on gastric motility during digestive tract. Methods Thirty - two rats were randomly divided into 4 groups (n = 8): PVN + MTL group, PVN + NS group, pituitary removal group + PVN + MTL group, pituitary Remove + PVN + NS group. The expression of MTL immunoreactive substance in the pituitary gland of rats was observed by fluorescence immunohistochemical staining. The changes of plasma MTL and intestine-migratory complex movement (MMC) in rats after pituitary gland removal were observed by using the method of gastric motility recording. RESULTS: (1) The frequency of MMC contraction in PVN + MTL group was significantly higher than that in PVN + MTL group [(4.9 ± 0.6) h / h vs [(11.2 ± 1.3) 5.3 ± 0.5) times / h] (P> 0.05); (2) MMC shrinking frequency significantly slowed down [(4.5 ± 0.8) times / h] Pituitary removal + PVN + MTL group MMC contraction frequency increased [(8.2 ± 0.7) times / h], compared with the control value was statistically significant (P <0.05); pituitary removal + PVN + NS group [(4.3 ± 0.9 ) Times / h] had no significant difference with the control (P> 0.05). (3) Immunofluorescence staining showed that there was expression of MTL immunopositive in pituitary gland. Conclusions Hypothalamus-pituitary plays a regulatory role in the interdigestive gastric motility.
