目的了解浙江省中东部地区白纹伊蚊种群中沃尔巴克氏体(Wolbachia)的感染状况及其基因型别,为蚊虫和蚊媒传染病的生物防制提供依据。方法用诱蚊灯法和人诱法采集浙江省中东部地区的白纹伊蚊雌性成蚊,PCR扩增沃尔巴克氏体的wsp基因,测序并进行系统进化分析。结果在6个白纹伊蚊采样点(杭州市、宁波市、嵊泗县、义乌市、丽水市和温州市)捕获白纹伊蚊雌性成蚊共192只,各个白纹伊蚊种群的沃尔巴克氏体核酸阳性率在25.0%~96.9%之间。获得11个wsp单倍型序列,序列长度在581~605 bp之间,碱基A、T、G、C的平均含量分别为31.4%、31.1%、21.5%和16.0%。各个wsp核酸序列的同源性在76.5%~99.8%之间,对应的氨基酸序列同源性在66.2%~100%之间。系统进化分析结果显示,11个单倍型序列被划分在2个超级组别(A组和B组)中,超级组别A属于w Alb A型沃尔巴克氏体,超级组别B属于w Alb B型沃尔巴克氏体。结论浙江省中东部白纹伊蚊普遍感染沃尔巴克氏体,其基因型为wAlbA和wAlbB型。
Objective To understand the infection status and genotypes of Wolbachia in Aedes albopictus from the eastern and eastern regions of Zhejiang Province and provide the basis for the biological control of mosquito and mosquito-borne diseases. Methods Female mosquitoes of Aedes albopictus were collected from mosquitoes and mosquitoes in the eastern and eastern regions of Zhejiang Province by using mosquito lamp method and human induced method. The wsp gene of Wolbachia was amplified by PCR and sequenced. Results 192 mosquitoes of Aedes albopictus were captured at six sites of Aedes albopictus (Hangzhou, Ningbo, Shengsi, Yiwu, Lishui and Wenzhou) The positive rate of the baculovirus nucleic acid was between 25.0% and 96.9%. Eleven wsp haplotypes were obtained. The length of the sequences was between 581-605 bp. The average contents of A, T, G and C were 31.4%, 31.1%, 21.5% and 16.0% respectively. The homology of each wsp nucleic acid sequence is between 76.5% and 99.8%, and the corresponding amino acid sequence homologies are between 66.2% and 100%. Phylogenetic analysis showed that eleven haplotypes were divided into two supergroups (group A and group B), supergroup A belonged to w Alb A type Wolbachia, super group B belonged to w Alb B type Wolbachia body. Conclusion The Aedes albopictus in central and eastern Zhejiang province is generally infected with Wolbachia, and its genotypes are wAlbA and wAlbB.
