目的研究超临界CO2萃取白芷挥发油最佳萃取工艺及其萃取物的化学成分。方法采用均匀设计法以萃取压力、萃取温度、萃取时间为因素,考察超临界CO2萃取白芷挥发油的最佳萃取工艺条件,并对萃取物进行GC×GC/TOFMS分析。结果最佳萃取工艺条件为:萃取压力17.8 MPa,萃取温度54℃,萃取时间175 min,此条件下的最大优化萃取率为4.17%。白芷萃取物中鉴定出86种化合物,相对质量分数在前3位的是香豆素类化合物,异氧化前胡素,别欧前胡素和氧化前胡素。结论采用均匀设计方法可以快速、准确地确定超临界CO2萃取技术萃取白芷挥发油最佳工艺条件,为白芷挥发油进一步开发利用提供实验依据;GC×GC/TOF MS分析对白芷挥发油化学成分研究提供了依据。
Objective To study the optimal extraction process of volatile oil from Angelica dahurica by supercritical CO2 and its chemical constituents. Methods The optimum extraction conditions of volatile oil extracted from Angelica dahurica by supercritical CO2 were investigated by uniform design with the factors of extraction pressure, extraction temperature and extraction time. The extracts were analyzed by GC × GC / TOF MS. Results The optimum extraction conditions were as follows: extraction pressure 17.8 MPa, extraction temperature 54 ℃, extraction time 175 min. Under these conditions, the maximum extraction rate was 4.17%. Angelica extract identified 86 kinds of compounds, the relative mass fraction of the top three are coumarins, isoxanthin, pre-imperatorin and oxidation of betanin. Conclusion The uniform design method can quickly and accurately determine the optimal extraction conditions of volatile oil from Angelica dahurica by supercritical CO2 extraction and provide experimental basis for the further development and utilization of volatile oil from Angelica dahurica. GC × GC / TOF MS analysis provides the basis for the chemical constituents of volatile oil from Angelica dahurica .
