采用氯胺-T法对胰岛素、牛血清白蛋白和猪超氧化物歧化酶进行125I标记,标记率采用淋洗分离时标记产品峰计数率与诸峰计数率之和的比值、三氯醋酸(TCA)沉淀、纸层析及纸电泳等四种方法进行测定。测定结果表明,第一种方法结果偏低,与后三种方法有显著差异,而后三种方法之间无显著差异,其中尤以TCA沉淀法操作简单、方便、速度快。还采用TCA沉淀法对上述几种125I标记产品的保存条件作了探讨,结果提示,标记产品加蛋白保护,并且在低温条件下或以冻干品形式存放时脱碘率最低。
Using chloramine-T method, insulin, bovine serum albumin and superoxide dismutase were labeled with 125I, the ratio of labeled product peak count rate to the sum of peak count rate was determined by elution. The ratio of trichloroacetic acid TCA) precipitation, paper chromatography and paper electrophoresis four methods were measured. The results show that the first method is low, with the latter three methods were significantly different, but no significant difference between the latter three methods, especially the TCA precipitation method is simple, convenient and fast. The TCA precipitation method was also used to investigate the storage conditions of the above 125I-labeled products. The results indicated that the labeled product was protected by protein and the deiodination rate was the lowest when stored at low temperature or in the form of lyophilized product.
