目的:观察基质金属蛋白酶-9(MMP-9)和高尔基体糖蛋白73(GP73)在原发性肝癌(primaryhepatocarcinoma,PHC)组织中的表达,并探讨其与原发性肝癌临床病理特征的关系。方法:应用免疫组织化学SP法检测55例原发性肝癌和15例正常肝组织中MMP-9和GP73的表达情况。结果:MMP-9和GP73在PHC组中的阳性表达率分别为74.5%、81.8%,均明显高于正常肝组织组(26.7%、33.3%),2组之间的差异显著(P<0.01)。GP73的阳性表达与患者的性别、年龄、肿瘤的数目及包膜有无无关(P>0.05),而与肿瘤的大小、Edmondson分级及有无转移有关(P<0.05);MMP-9在肝癌组织中的表达与患者的性别、年龄、肿瘤的数目无关(P>0.05),与肿瘤的大小、包膜有无、Edmondson分级及有无转移有关(P<0.05)。结论:MMP-9和GP73在肝癌组织中的表达升高,二者联合检测可作为PHC诊断和预后的指标。
OBJECTIVE: To investigate the expression of MMP-9 and GP73 in primary hepatocarcinoma (PHC) tissues and to investigate its relationship with the clinicopathological features of primary hepatocellular carcinoma . Methods: The expressions of MMP-9 and GP73 in 55 cases of primary hepatocellular carcinoma and 15 cases of normal liver tissue were detected by immunohistochemical SP method. Results: The positive rates of MMP-9 and GP73 in PHC group were 74.5% and 81.8%, respectively, which were significantly higher than those in normal liver tissue (26.7% and 33.3%, P <0.01 ). The positive expression of GP73 was not associated with the gender, age, tumor number and the presence or absence of the capsule (P> 0.05), but was related to tumor size, Edmondson grade and metastasis (P <0.05) Tissue expression was not related to the patient’s gender, age, number of tumors (P> 0.05), and the tumor size, presence or absence of coating, Edmondson grade and metastasis (P <0.05). Conclusion: The expression of MMP-9 and GP73 in hepatocellular carcinoma tissue is increased. The combined detection of MMP-9 and GP73 may be used as an index for the diagnosis and prognosis of PHC.
