用紫外分光光度法测定蛋白胨总氮是一种新的测定方法(简称紫法)。我们对该法做了大量实验。在实验过程中,我们发现《紫法》一文中的试剂用量及实验条件均存在一定的问题,诗写此文,以与祝平乾同志商榷。 1 关于K_2S_2O_8的用量,《紫法》中规定为加入K_2S_2O_8(0.1mol.L~(-1))20ml。我们在实验中发现,该用量与实验实际需要量相差甚远,实验需要加入10g固体K_2S_2O_8。 0.2g蛋白胨中含氮约32mmol,约合2.3mmolN,N转化为NO_3~-时,N与O的摩尔比为1
Determination of total peptone by UV spectrophotometry is a new assay (referred to as the purple method). We have done a great deal of experimentation with this law. During the experiment, we found that there are some problems in reagent amount and experimental conditions in the article “Purple Method”, and the poem is written to discuss with Comrade Zhu Ping. 1 About the amount of K 2 S 2 O 8, “Purple Act” provides for adding K_2S_2O_8 (0.1mol.L ~ (-1)) 20ml. We found in the experiment, the amount and the actual needs of the experiment a far cry from the experiment need to add 10g of solid K_2S_2O_8. 0.2g peptone contains about 32mmol nitrogen, about 2.3mmolN, N is converted to NO_3 ~ -, N and O molar ratio of 1
