目的:探讨低氧和SD大鼠阴茎海绵体平滑肌细胞(CCSMC)凋亡的关系。方法:体外培养CCSMC,免疫组化鉴定细胞;低氧(1%O2浓度)干预12、24、48、72 h,常规氧浓度作为对照,流式细胞术测定细胞周期变化和凋亡情况。结果:体外培养的CCSMC生长良好,抗平滑肌α-肌动蛋白单克隆抗体免疫组化DAB法染色阳性。流式细胞术检测CCSMC G0/G1期在48 h内细胞比例逐渐增加,后下降;S期细胞比例与G0/G1期呈相反趋势;G2/M期细胞比例无明显规律。结论:CCSMC在低氧环境下,随时间的延长凋亡程度加重,48 h达到最大值,进一步延长时间细胞裂解,不能加重凋亡。
Objective: To investigate the relationship between hypoxia and apoptosis of corpus cavernosum smooth muscle cells (CCSMCs) in SD rats. Methods: CCSMCs were cultured in vitro and identified by immunohistochemistry. Hypoxia (1% O2 concentration) for 12, 24, 48 and 72 h were used as control. The cell cycle and apoptosis were determined by flow cytometry. Results: The CCSMC cultured in vitro grew well and anti-smooth muscle α-actin monoclonal antibody was positive for immunohistochemical DAB staining. The proportion of cells in G0 / G1 phase of CCSMC increased gradually and then decreased in G0 / G1 phase. The percentage of cells in S phase was opposite to that in G0 / G1 phase. The proportion of cells in G2 / M phase had no obvious regulation. CONCLUSION: Under hypoxic conditions, CCSMC increased the degree of apoptosis with time and reached its peak at 48 hours, further prolonging the time of cell lysis and not increasing apoptosis.
