[目的]探讨龟板含药血清对骨髓间充质干细胞(MSCs)肝细胞生长因子受体(c-met)表达及迁移的影响。[方法]密度梯度离心法分离、培养MSCs,改良boydon小室法检测MSCs迁移功能,Western blot检测c-met蛋白表达,携带siRNA腺病毒转染下调c-met表达。[结果]龟板含药血清共培养明显增强MSCs的迁移能力,且呈浓度依赖性。同时,龟板含药血清增加MSCs的c-met蛋白表达,低表达c-met蛋白可明显减弱龟板含药血清的促MSCs迁移作用。[结论]龟板含药血清可能通过肝细胞生长因子(HGF)/c-met轴促MSCs迁移。
[Objective] To investigate the effect of turtle plate containing serum on the expression and migration of hepatocyte growth factor receptor (c-met) in bone marrow mesenchymal stem cells (MSCs). [Methods] MSCs were isolated and cultured by density gradient centrifugation. The migration of MSCs was detected by modified boydon chamber method. The expression of c-met protein was detected by Western blot. The expression of c-met was induced by transfection with siRNA adenovirus. [Result] Coagulation of turtle shell containing serum significantly enhanced the migration ability of MSCs in a concentration-dependent manner. At the same time, the turtle plate containing serum increased the c-met protein expression of MSCs, low expression of c-met protein can significantly reduce turtle plate containing serum-induced migration of MSCs. [Conclusion] The medicated serum of turtle shell may migrate the MSCs through the hepatocyte growth factor (HGF) / c-met axis.
