目的探讨颐脑解郁复方对脑卒中后抑郁(PSD)大鼠行为学和海马CA1区病理损伤的干预作用。方法将168只SPF级SD大鼠随机分为正常组、假手术组、卒中组、PSD组、西药组和中药组,正常组、假手术组各24只,其余组各30只。正常组不干预,假手术组不插入线栓,卒中组仅行大脑中动脉阻塞手术,PSD组、西药组、中药组卒中术后1周应用慢性束缚应激1周复合单笼饲养法制备PSD模型。造模开始时,西药组灌胃盐酸氟西汀,中药组灌胃颐脑解郁复方,其余各组灌胃蒸馏水,每日1次。第2、4、8周末,各组大鼠进行旷场试验等行为学测试,HE染色观察海马CA1区病理变化。结果除第2周外,同一时间点中药组大鼠各项行为学评分均高于PSD组。同一时间点中药组大鼠海马CA1区较PSD组结构完整,细胞排列整齐,形态正常。结论颐脑解郁复方可改善大鼠PSD症状,并对海马CA1区具有保护作用。
Objective To investigate the effect of Yinao Jieyu Compound on the behavior and the pathological damage of hippocampal CA1 in post-stroke depression (PSD) rats. Methods 168 SPF SD rats were randomly divided into normal group, sham operation group, stroke group, PSD group, western medicine group and traditional Chinese medicine group, normal group and sham operation group, 24 rats in each group. The normal group did not intervene, the sham-operated group did not insert the thrombus, the stroke group only received the middle cerebral artery occlusion operation. The PSD group, western medicine group and traditional Chinese medicine group were prepared by chronic restraint stress 1 week combined with single-cage feeding model. At the beginning of model establishment, the western medicine group was given fluoxetine hydrochloride, while the Chinese medicine group was given gavage Yiyou Jieyu compound, and the other groups were given distilled water once a day. At the end of 2, 4, and 8 weeks, rats in each group were subjected to behavioral tests such as open-field test. The pathological changes of hippocampal CA1 region were observed by HE staining. Results Except for the second week, all the behavioral scores of the rats in the TCM group were higher than those in the PSD group at the same time point. At the same time point, the hippocampal CA1 region of rats in the traditional Chinese medicine group was more complete than the PSD group, and the cells were arranged neatly with normal morphology. Conclusion Yinao Jieyu Compound can improve the symptoms of PSD in rats and protect the hippocampal CA1 region.
