樟芝(Taiwanofungus camphoratus)发酵液和经95%乙醇浸提后的菌丝体醇提物分别用石油醚和氯仿进行分级提取,得到石油醚和氯仿提取物。对不同提取物进行体外抑制SPCA-1肺癌细胞增殖实验,发现各提取物均可抑制SPCA-1细胞的增殖,其中发酵液石油醚提取物抑制作用最好,IC50为62.5μg/mL,发酵液和菌丝体氯仿提取物的抑制作用较好,IC50分别为120.9和109.5μg/mL,菌丝体石油醚提取物的抑制作用较差,在25~150μg/mL作用浓度下,抑制率低于20%。各提取物对SPCA-1细胞凋亡和生长周期作用的测定结果表明,各提取物在肺癌细胞SPCA-1中通过诱导细胞凋亡及S期周期阻滞来表现其抑制作用。
The extract of Taiwanofungus camphoratus and the mycelial alcohol extract after 95% ethanol extraction were fractionated by petroleum ether and chloroform, respectively, to obtain petroleum ether and chloroform extract. In vitro experiments on proliferation of SPCA-1 lung cancer cells by different extracts showed that all the extracts could inhibit the proliferation of SPCA-1 cells. Among them, petroleum ether extract of fermentation broth had the best inhibitory effect with IC50 of 62.5 μg / mL, And mycelium chloroform extract showed better inhibitory effect with IC50 of 120.9 and 109.5 μg / mL, respectively. The mycelial extracts of petroleum ether had less inhibitory effect. Under the concentration of 25 ~ 150μg / mL, the inhibitory rate was lower than 20%. The results of the determination of the effect of each extract on the apoptosis and the growth cycle of SPCA-1 cells showed that each extract exhibited inhibition by inducing apoptosis and S phase arrest in the lung cancer cell SPCA-1.
