【目的】本研究针对痛风与高尿酸血症形成的原因,运用动物模型探讨阿姆热提取物防治痛风与高尿酸血症的作用机理。【方法】采用小鼠灌胃腺嘌呤加乙胺丁醇造成小鼠高尿酸血症模型,以别嘌呤醇为阳性对照药,观察阿姆热药物的醇提取物与水提取物对高尿酸血症小鼠血清尿酸水平及肝脏黄嘌吟氧化酶的影响;采用大鼠自身对照方法,以苯溴马隆为阳性对照药,分别用阿姆热药物的不同提取物及不同剂量连续灌胃给药三天,收集灌胃给药后6小时的尿液,测定尿酸和尿量,通过排泄实验评价该药的促尿酸排泄作用;选择鹌鹑作为模型动物,以别嘌呤醇为阳性对照药,将酵母加黄豆粉作为造模剂,制备与人类代谢类似的高尿酸血症动物模型,观察阿姆热水提物对鹌鹑血清尿酸、血糖、甘油三酯的影响;对实验小鼠的肾脏和鹌鹑的肝、脾、肺、肾进行病理检查,观察其形态学改变。【结果】阿姆热醇提物给药14天,血UA未降低,反而升高非常显著(P<0.01);给药21天,血UA下降,且高剂量组降尿酸作用明显(P<0.05),但阿姆热醇提药并没有影响肝脏XOD活性;阿姆热水提物给药14天,血UA即下降,高剂量组血UA的降低有统计学意义(P<0.05),给药21天,高剂量与低剂量给药组血UA均显著下降(P<0.01),且接近正常水平,对肝脏XOD活性均有抑制作用(P<0.05)。别嘌呤醇给药组给药14天,血尿酸即下降(P<0.05),给药21天血尿酸降低更显著(P<0.01),对肝脏XOD有抑制作用(P<0.05)。与模型组比较,给药组肾脏病变改善。阿姆热醇提物没有使正常大鼠尿酸排泄增加,其高剂量组甚至在给药第二天、第三天尿酸排泄减少(P<0.05);阿姆热水提物低剂量组在给药第二天出现促尿酸排泄(P<0.05),高剂量组与苯溴马隆组在给药第一天尿酸排泄即增加(P<0.05)。用酵母加黄豆粉造模,模型组血尿酸较正常对照组升高(P<0.05);酵母加黄豆粉加双氯芬酸组的血UA也较正常对照组升高(P<0.05),尽管酵母加黄豆粉组与酵母加黄豆粉加双氯芬酸组比较无统计学差异,但后者血UA偏低;造模的同时给阿姆热水提物,高剂量与低剂量给药后均可使血UA下降(P<0.05),且高剂量组血UA较低。别嘌呤醇给药组血尿酸也较模型组降低(P<0.05)。阿姆热水提物对鹌鹑血糖(Glu)与血甘油三酯(TG)影响不明显。观察各组鹌鹑内脏的病理形态改变,发现正常对照组、模型组、各给药组之间未见明显的差别。【结论】阿姆热醇提物长期给药有降尿酸作用,但没有证据表明其是通过抑制肝脏XOD活性与促尿酸排泄作用,具体机制有待探讨;阿姆热水提物降尿酸作用明显,通过抑制肝脏XOD活性与促尿酸排泄作用使血UA下降。各给药组肾脏的病理改变明显轻于模型组。用酵母造模较腺嘌呤造模对动物内脏的损害小。上述研究结果表明,阿姆热可用于防治痛风、高尿酸血症及其并发症,且其水提物具有机制明确、降尿酸显著的优点,值得进一步研究开发。【Objective】The purpose of the present study is to explore the mechanism of a folk plant Amure extracts on prevention and therapy of gout and hyperuricemia by using animals models.【Methods】Hyperuricemia model was induced by introgastric administration to mice with Adenine and Ethambutol.The effects of Amure ethanol and water extracts on the level of uric acid in plasma and the inhibitory activity on liver xanthine oxidase(XOD) were observed with allopurinol as a positive control. The model for evaluating effectivity of promoting excretion of uric acid was established by oral administrating benzbromarone to rats with self control. The contents of uric acid and urine volume were determinated continuously in three days six hours after administrating with different doses of the two extracts. Hyperuricemia model in quails was made by feeding yeast and soybean flour.
