Besides hypotheses that cancer stem cells (CSCs), representing a rare population of cancer cells exhibiting stem cell features such as self-renewal, differentiation, tissue regeneration and drug resistance, may originate from adult stem cells or committed progenitor cells it is also assumed that the fusion between stem cells and somatic cells and cancer cells, respectively, could give rise to a CSC phenotype.To investigate for the fusogenic potential of human breast stem cells and human breast cancer cells we co-cultured human M13SV1-EGF-Neo breast stem cells with HS578T-Hyg and MDA-MB-435-Hyg, respectively, human breast cancer cells.After 24h, cells were cultured under selective conditions (+ G418, + Hyg) and cell clones emerged from spontaneous fusion events were isolated and cultivated separately.Short tandem repeat (STR) analysis of hybrid cells showed an overlap of the parental alleles indicating that hybrid cells have been originated from real cell fusion events.
