Alternative polyadenylation (APA) is a widespread phenomenon that generates transcript isoforms with alternative 3UTR.Through alteration of 3UTRs, APA potentially regulates the stability, cellular localization and translation efficiency of target RNAs.Previous work in our lab demonstrated that mei-P26 has alternative 3UTR in fly testes, longer in tut (tumorous testis), barn, or bgcn mutant arrested at spermatogonia stage while shorter in wild type.We aim to explore the relationship between the change of 3UTR and germline differentiation, and how differentiation-related 3UTR is regulated.By Poly(A) Site Sequencing (PAS-Seq),we found that hundreds of genes 3UTR were lengthened in tut, bam, or bgcn mutant testes.GO term analysis showed that the genes with alternative 3UTR in these mutant testes are associated with RNA regulation and germ cell development.Our data indicate that 3UTR length is mis-regulated in spermatogonia-enriched mutant testes, suggesting that APA may be involved in germ cell differentiation.
