  Gene therapy is a method designed to express the therapeutic proteins in target cell.Compared with the traditional therapeutic methods such as surgery,chemotherapy and radiotherapy,it has greater potential to inhibit cancer.Since the intracellular transport of gene vector/plasmid DNA is an important part of gene therapy,its mechanistic understanding is of extreme importance.The targeting gene vector P123-PE1-R13 has been successfully made in our previous work,P123-PEI-R13/DNA complex exhibited higher transfection efficiency and lower cytotoxicity.In order to investigate its intracellular transport,chemical inhibitors specific to the various intracellular pathways have been applied.The results revealed that the cellular uptake of the P123-PEI-R13/DNA complex was conducted simultaneously via clathrin-mediated endocytosis,caveolin-mediated endocytosis,macropinocytosis and a possible energy-independent pathway.
