The effects of SYK on proliferation and lactation in mammary epithelial cells of dairy cows

  Spleen tyrosine kinase (SYK) is a non-receptor tyrosine kinase that is expressed most in hematopoietic cells.It has been considered a cell-specific signal transducer involved in cell proliferation and differentiation.However, the biological role of SYK in dairy cow mammary gland remains largely unknown.Here we showed that SYK was expressed in mammary epithelial cells of dairy cows.MTT results showed that overexpression of SYK enhanced mammary epithelial cell proliferation, whereas knockdown of SYK had the opposite effects.The overexpression of SYK in cultured dairy cow mammary epithelial cells markedly increased the protein levels of β-casein, AKT1,pAKT1, MAPK, pMAPK, and STAT5, whereas knockdown of SYK decreased the protein levels of β-casein, AKT1, pAKT1, MAPK, pMAPK, and STAT5.Overall these results reveal that SYK is crucial in mediating mammary epithelial cell proliferation and lactation in mammary glands of dairy cows.
