  Aims To evaluate and compare the effect of total body irradiation (TBI) with busulfan/cyclophosphamide (BU/CY) as conditioning regimen on the development of hepatic veno-occlusive disease (HVOD) occurring after hematopoietic stem cell transplantation (HSCT).Methods Mice received either TBI (7.5 Gy with rate at 0.67 Gy/min) (n=20) or BU/CY (80 and 200mg/kg/d)(7-4 days for BU and 3-2 days for CY before transplantation) (n=20) followed by HSCT.Liver and whole blood collected 15 days post HSCT were used for the analysis of liver pathology changes by H&E, immunohistochemical and Mason staining, ultrastructure by transmission electron microscope, function (level of alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP)) by Bio-Chemical Analyzer, peripheral white blood cell (PWBC) count by blood cell counter, reticulocytes and platelet activation by flow cytometry.
