A gene-specific microsphere suspension array coupled with1 5-plex PCR was developed to screen bacterial samples rapidly for ten strains ofbacteria:Shigella spp(S.Flexneri,S.Dysenteriae,and S.Sonnei),Staphylococcusaureus,Vibrio cholerae(serology 01 and 0139),Legionella pneumophila,andClostridium botulinum(types A,B,and E).Fifteen sets of highly validatedpfimem were chosen to amplify target genes simultaneously.Correspondingoligonucleotide probes directly conjugated with microsphere sets were used tospecifically identify PCR amplicons.Sensitivity tests revealed that the arraycoupled with single PCR was able to detect purified genomic DNA atconcentrations as low as 10 fg/μl,corresponding to less than 10 cells,while themultiplex detection limit was 1 0 fg/μl-10 pg/μl.The suspension array with15-plex PCR was also validated the utility for simulated samples and realsamples.
