Organometallic Ru(Ⅱ)/Os(Ⅱ)arene complexes of the type [(η6-arene)Ru(en)Cl]+,[1] where the arene = e.g.p-cymene,biphenyl(bip)or tetrahydroanthracene(tha)and en = ethylenediamine,are toxic to cancer cells,including cisplatin-resistant cell lines.Our results[1,2] show that there two kinds of intercalation of Ru-arene into DNA,including classic and a novel penetrating intercalation.We also relate these dual mode interactions(coordination and intercalation)to the mechanism of anticancer activity and to structure-activity relationships.In addition,we find that the interactions between these complexes and DNA show close similarities to those of covalent polycyclic aromatic carcinogens.
