We reported the development of two- and three-input logic devices based on the fact that Hg2+-,Ag+-,and/or coralyne can drive fluorescence quenching of oligonucleotide-based probe.The designed probe is a single-stranded oligonucleotide that contains a a carboxyfluorescein (FAM) at the 5-end and a 4-([4-(dimethylamino)phenoyl]azo)-benzoic acid (DABCYL) at the 3-end.Becuase a single-stranded oligonucleotide consists of a series of 58 random DNA bases (DNA58,GTC TCT GTG TGC GCC AGA GAA CAC TGG GGC AGA TAT GGG CCA GCA CAG AAT GAG GCC C),Hg2+-,Ag+-,and coralyne can force the conformation change of the DNA58 from a random coil to a folded structure via the coordination of thymine-Hg2+- thymine,cytosine-Ag+-cytosine,and adenosine2-coralyne-adenosine2,respectively.This conformational change induced direct contact between FAM and DABCYL,causing collisional quenching of fluorescence.The addition of thiol-containing molecules and chelating agents to a solution of the Hg2+-DNA58 complexes resulted in the restoration of the fluorescence of FAM.Similar fluorescence response was observed in the case of the Ag+-DNA58 complexes.Moreover,the fluorescence of the coralyne-DNA58 complexes lighted up in the presence of heparin.According to the abovementioned results,a set of logic gates involving two-input and three-input systems was demonstrated,including OR,AND,NOR,INH,IMPLICATION and three-input NOR gate.These results displays the higher order circuts for logic gate operation based on oligonucleotide-based probe.
