  OBJECTIVE:To assess the effect and dose-response relationships of Tanreqing injection on Lipopolysacchaide(LPS)induced airway mucus hypersecretion in HBE16 airway cells.METHODS:To explore the inflammation model of HBE16 cells by LPS with optimal processing time and concentration by CCK-8 method;to explore the maximum concentration of TRQ and baicalin for HBE16 cells by CCK-8 method;according to the maximum concentration of TRQ and baicalin for HBE16 cells,the groups were divided into control group,LPS group,LPS plus drug intervention group(TRQ:1:32,1:64,1:128,1:256,1:512;baicalin:10 μg/ml、50 μg/ml、100 μg/ml);the secretions of MUC5AC of HBE16 cells treated with LPS plus TRQ or baicalin in cell culture supernatants were quantified by enzyme-linked immunosorbent assay(ELISA).Tthe messenger RNA(mRNA)expressions of MUC5AC of HBE16 cells treated with LPS plus TRQ or baicalin in cell RNA were tested by quantitative real-time polymerase chain reaction(real-time RT-PCR).The expression of phospho-IKKα/β,phospho-NF-κB p65 and phospho-IκB-α of HBE16 cells treated with LPS plus TRQ or baicalin for 24 hours were tested by Western blotting to determine whether the signaling pathway NF-κB was involved in the anti-inflammatory action of TRQ and baicalin.
