Objective: To investigate the superiority and feasibility of chitosan thermosensitive hydrogel with Epidermal Growth Factor (EGF) sustained-release properties.Methods: Using tissue explant in vitro primary culture 7d-old SD rat submandibular gland cells (RSMGCs), identify the cell origin by immunofluorescence and passaged.To prepare chitosan thermosensitive hydrogel by neutralization reaction occurred in chitosan andβ-glycerolphosphate.Observe its general characteristics, ultrastructure and sustained released properties for EGF.Respectively use EGF sustained released chitosan thermosensitive hydrogel, free EGF, chitosan thermosensitive hydrogel and culture medium as experimental group1 and 2,control group and blank group, co-cultured with the first two grown well generation of RSMGCs.Detect the proliferation and secretion of cells by MTT and lodine-amylase colorimetric.Use concentration and time factor for multi-level single factor analysis of variance pairwise comparison by SPSS 13.0.Test levelα=0.05,P<0.05 considered significant difference.Using Acridine orange(AO) immunofluorescence staining and laser scanning confocal microscope (LSCM) to observe cell viability after co-cultured.
