Human erythropoietin (hEPO), an endogenously produced glycoprotein, plays a fundamental role in erythropoiesis controlling the formation of red blood cells.Production of recombinant human erythropoietin (rhEPO) by molecular biology techniques has made possible its abuse in competitive sports, so there needs a high sensitivity method to detect rhEPO.This paper developed a method for the detection ofrhEPO by capillary electrophoresis with laser-induced fluorescence (CE-LIF).First, rhEPO was denatured by heating at 98℃ for 8 minutes, and then derivatized with 3-(2-furoyl)quinoline-2carboxaldehyde (FQ) at 65℃ and analyzed using CE-LIF.We used carboxymethyl chitosan dynamic coating to prevent the adsorption of rhEPO on the wall of bare fused-silica capillary.The results of CE-LIF were compared with that of CE-UV, demonstrating that the sensitivity of LIF detection was much higher than that of UV detection(Fig.l).However, we could not get the peaks of different glyeoforms of derivative EPO by LIF as by UV detection.
