The H19 is one of the most abundantly expressed genes during mammalian embryonic and fetal development. It was found to be paternally imprinted in some mammalian species, such as human, mouse, sheep, pig and cattle. However, the structure, expression and imprinting status of goat H19 gene remain unknown. In present study, a goat H19 fragment of 1139bp, which covers most part of this gene, including full-length of exon 3, exon 4, intron 2, intron 3 and intron 4 and partial sequences of exon 2 and exon 5, was obtained. The obtained part of goat H19 has a high identity of 96.2% and 90.5% with the corresponding region of ovine H19 and bovine H19, respectively. The H19 was expressed in most tissues of goat fetuses and in the liver, lung, pancreas, longbsimus dorsi muscle, spleen and kidney of adult goats. A polymorphism in mature RNA was identified at goat H19 exon 5, and by analyzing the genotypes of the parents, the offspring and the expressed allele in tissues of the offspring at mis polymorphic site, we found that H19 was paternally imprinted. The results indicated that molecular characteristics and imprinting status of goat H19 was conserved compared to other mammalian species.
