　　Type Ⅲ restriction enzymes are ATP dependent enzymes composed of restriction and modification subunits.The holoenzyme is a heterotetramer.To introduce one double strand break 25-27 bp downstream of recognition site,the presence of two oppositely oriented recognition sites on circular DNA is essential,using the type Ⅲ restriction enzyme EcoP15I.
