Introduction ThereareonlyfewabouttherelationshipsofthemethodsofpreparingPRPandthecellcomponentsandsecretiontimes. Thepurposeisanalysisofthecomponents,amountofgrowthfactors(GF),thesecretionmechanicsusingthePRPwhichprepared by2kindsofkit(PRPHPandPRPLP). Material and Methods Each1x106/ulof9samplesofPRPhadbeencultured20daysafterhandledbyHumanDiploidFibroblasts(HDFs).Duringthe culture,weharvestedmediasupernatants11timesatculturetime30min,1hour,1,2,4,6,9,10,15,20days.Weanalyzedthe quantityoftheGFsandsecretionmechanics. Atbloodcomponents,wemeasuredthequantityofplateletandWBC.AtGFs,wemeasuredthequantityofGFsandcytokines(IL1-B,PDGF-AA,PDGF-ABBB,VEGF,MMP-13,TGF-B1,TGF-B2,TGF-B3,IGF-1,IGFBP-1,IL-18,andTNF-A)andthesecretion mechanics. Results TheconcentrateofplateletofPRPgroupsishigherthanwholeblood.Notonlytheconcentrationofplateletbutalsothe concentrationofWBCofPRPHPishigher.TheconcentrateofWBCofPRPLPisthelowest,thoughtheconcentrateofplateletis higherthanwholeblood. Also,concentratesoftheIL-1B,TNF-AwhichareinflammatorycytokinesofPRPgroupsislowerthanwholeblood. RegardingGFsecretionmechanics,PDGF-AAandPDGF-ABBBstartedtobesecretedasamaximumimmediatelyafterapplication ofPRP,anddecreaseduntiltheday4.VEGFandTGF-B1,2startedtoincreaseaftertheday2andcontinuedtoincreaseuntilthe day20.CytokineIL-18startedtoincreaseafteranhour,andTNF-Aalsostartedtoincreasegraduallyafteranhour.IGFBP-1 increasedgraduallyinthreegroups,andsecretionratewassubstantiallyhighinPRPHP. Conclusion Theconcentrationofplatelet,WBCandGFsdifferedaccordingtomethodsofplateletproduction.Concentrationofplateletand GFswashigherinPRPgroupthanwholeblood. Especially,secretionmechanicsdifferedaccordingtoplateletproductionmethods. Inconclusion,weconsiderconcentrationofplatelet,WBC,GFsinadditiontoGFssecretionmechanicstobeimportantwhenclinical doctorsusePRPtreatment.
